Effect of Growth Regulators on Cell Growth , Embryogenesis and Regeneration of Plantlets from Carrot (Daucus carota L.) Cell and Callus Culture



Abstract
The effects of various growth regulators, such as picloram, kinetin, 2, 4-D, and ABA, on the cell growth, embryogenesis, and regeneration of plantlets from embryoids formed through cell suspension and callus culture of mature carrot roots were investigated in this experiments.
The addition of 0.1§·/§¤ picloram to hatch suspension culture media resulted in earlier cell devision and better cell proliferation than that of 0.3§·/§¤ 2, 4-D or kinetin. However, the addition of picloram or kinetin was inhibitory to the embryogenesis.
ABA treatment, conc. from 0.3 to 10§·/§¤, increased the number of young embryos, especially at latter concentration, numerous young and normal embryos were observed. The callus growth was promoted with 0.3§·/§¤ ABA treatment compared with control.
The different forms of plantlet regeneration from embryoids were observed, depending on the number of times subculture employed. Normal differentiation with initial culture, but with passing subculture times, callus formation of enlarged roots and of shoot apex with elongated roots, and multiple shoots from a embryoid were observed. ABA treatment had effectively reduced the frequencies of abnormal embryos, resulting in high frequencies of normal plantlets.
